Structure and Modifications

SNAP-8 is an octapeptide composed of glutamyl and heptapeptide amino acids. Its structure allows it to mimic the N-terminus of SNAP-25, a protein involved in the SNARE complex, which is essential for neurotransmitter release and muscle contraction.

Synthesis and Studies

SNAP-8 has been synthesized using a variety of methods, including solid phase synthesis. Studies have shown that it is effective in reducing the depth of facial wrinkles caused by muscle contraction, especially on the forehead and around the eyes.

Mechanism of Action

SNAP-8 modulates the formation of the SNARE complex involved in muscle stimulation by mimicking SNAP-25. It acts as a competitor to SNAP-25, taking up a position in the SNARE complex, interfering with its formation and reducing stability, thus relaxing the muscles. This mechanism is similar but different to that of Botox, which completely blocks the formation of SNAREs.

The synthesis of SNAP-8 involves a final freeze-drying step. The freeze-dried product is usually obtained in the form of polymorphic crystalline powder, which means that some aggregates and crystal size differences may occur locally. This polymorphism is not related to chemical differences, and the extensive work carried out by the analytical department ensures the homogeneity of the product. SNAP-8 is available in the form of SNAP-8 powder (a powdered octapeptide that is easily soluble in water) or SNAP-8 solution (a water solution containing 0.5g/L of the powdered version). It can be added at the final stage of manufacturing the product, provided that the temperature is below 40 degrees Celsius. Considering the concentration of the peptide in the SNAP-8 solution, it is recommended that 3% to 10% of the solution be present in the final formulation to obtain significant anti-wrinkle activity.

Storage and Shelf Life

SNAP-8 powder and SNAP-8 solution must be stored in a cool, dark and clean place to ensure a shelf life of at least twelve months. For long-term storage, it is recommended that both forms be stored at 40C, which can extend the shelf life to at least - eight months. In rare cases, refrigerated storage of SNAP-8 solution can lead to precipitation of the preservative. This does not affect the integrity of the product.
